I think it largely depends on what you're applying for. When I'm participating on a search committee, it means a lot for someone to tailor their resume to the posting and write a cover letter that clearly indicates what skills, experience, or qualifications they are bringing to the table and how they might apply to the specific job. To me, this sets them apart from a generic resume or a generic cover letter. This is especially helpful if you are applying for a position that is more of a slant fit or the way that someone "fits" into a position may not be as obvious just from the resume alone.
